**Experimental Principle:** The assay is based on the double-antibody sandwich ELISA method. A microtiter plate is pre-coated with purified human midkine (MK) antibody to serve as the solid-phase capture antibody. After adding the sample, any MK present will bind to the immobilized antibody. A horseradish peroxidase (HRP)-labeled MK antibody is then added, forming an antibody-antigen-enzyme-labeled antibody complex. Following a thorough wash step, the TMB substrate is introduced. Under HRP catalysis, TMB turns blue, and the addition of stop solution changes the color to yellow. The intensity of the yellow color is directly proportional to the MK concentration in the sample. Absorbance is measured at 450 nm using a microplate reader, and the MK concentration is determined by comparing the sample OD values to a standard curve.
