**Purpose of Use:** This kit is designed for the quantitative determination of Substance P (SP) in human serum, plasma, and other related biological samples. It provides a reliable and efficient method to measure SP levels, which can be useful in clinical research and diagnostic applications.
**Experimental Principle:** The kit utilizes the double-antibody sandwich ELISA method to detect the concentration of human Substance P in the sample. A microtiter plate is pre-coated with a specific monoclonal antibody against SP. After adding the sample, SP in the sample binds to the immobilized antibody. Then, a horseradish peroxidase (HRP)-labeled secondary antibody is added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following a thorough washing step, TMB substrate is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow upon addition of the stop solution. The intensity of the color is directly proportional to the amount of SP present in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the SP concentration is calculated based on a standard curve.
