Objective: To explore the preventive effect and mechanism of magnesium valproate on acute cerebral ischemia-reperfusion injury in mice. Methods: Mice were randomly divided into sham operation group, model group and magnesium valproate high and low dose (0.04, 0.02 mg · g-1) group, 10 mice in each group, and the corresponding drugs were given by gavage twice a day , For 14 consecutive days, 1 hour after the last administration, the model of acute cerebral ischemia-reperfusion injury was established in the latter three groups of mice. After successful modeling, the brain index and the content of malondialdehyde (MDA) in the brain tissue of each group of mice were detected. Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity, and observe the changes in cell morphology of cerebral cortex and hippocampus CA1 area. Results: Compared with the model group, mice with high and low doses of magnesium valproate alleviated neuronal damage, brain index and MDA content were significantly reduced, GSH-Px activity was significantly increased (P <0.01 or P <0.05), and C The SOD activity of the magnesium valerate high-dose group was significantly increased (P <0.01). Conclusion: Magnesium valproate has a preventive effect on acute cerebral ischemia-reperfusion injury in mice, and its mechanism may be related to the antioxidant effect.
